The goal of my laboratory is to understand how post-translational modifications including complex carbohydrates (glycans) contribute to biological function. Our lab is unlike any other in the world; we are the only lab that combines the three orthogonal approaches required to define structure/function relationships in human glycoproteins: 1) glycoproteomics of purified proteins from primary human tissue to define the modifications present in the human body, 2) structural biology approaches to measure conformationally heterogeneous glycoproteins, including solution nuclear magnetic resonance spectroscopy, x-ray crystallography and molecular modeling, and 3) biochemical measurements of glycoprotein function, including glycoprotein expression using human HEK293F cells and post-purification enzymatic remodeling of the glycans to achieve defined glycoforms.
